El sustrato de la fosfatasa se ha utilizado como sustrato en el ensayo de la fosfatasa ácida resistente al tartrato (TRAP) y la fosfatasa alcalina (ALP).
Sustrato cromógeno para usar en procedimientos ELISA de fosfatasa ácida y la alcalina. Rinde un producto final soluble que es de color amarillo y puede leerse espectrofotométricamente a 405 nm. La reacción de pNPP puede detenerse con NaOH 3 N y leerse a 405 nm.
